Job Description
The role
We are seeking a Specialist Research Technician to join a team that will be working on a recently awarded Cancer Research UK (CRUK) Discovery Programme. The aim of this research programme is to identify the antigens targeted by the alloreactive T-cell response in patients following stem cell transplantation for the treatment of Acute Myeloid Leukaemia (AML). Stem cell transplants given to patients contain alloreactive T-cells which come from the donor. These alloreactive donor T-cells can recognize and kill leukemic cells within the patient which is how the stem cell transplant is able to cure their cancer. However, in a proportion of patients, donor T-cells can fail to kill leukemic cells resulting in relapse or can attack normal tissues resulting in unpleasant side effects which are sometimes fatal. This research programme will utilise an antigen discovery approach to identify the antigens targeted by alloreactive donor T-cells. The aim of this research is to generate data that will enable the future design of strategies to improve clinical outcomes following stem cell transplantation.
What will you be doing?
The role holder will provide technical support for research activities being undertaken within the Clinical T-cell Immunology research group which is led by Professor Linda Wooldridge. The research group is located within the Biomedical Sciences Building.
Responsibilities of the role will include:
- Handling and processing of human biological samples, to include sample receipt, logging storage and maintenance of records in compliance with the Human Tissue Act
- Isolation of T-cells from patient samples after stem cell transplantation
- The culture of T-cells and mammalian cell lines
- T-cell effector function assays and antigen discovery techniques
- Running flow cytometry staining and analysis
- Manufacture of soluble proteins such as pMHCI monomers
